It is possible to have a palette of approximately 500 colors and can convert an image 24-bit in a palette of 40 colors optimized?

I want to buy the current version, but this time the next version 5?

You can use the Quantize8Bpp() method of the Imaging class:

The next V5 major release will be available within 1 - 3 month.

If you purchase the actual V4 edition you will get the V5 for free.
